7-Hydroxymethotrexate是甲氨蝶呤的I期代谢物，由肝醛氧化酶转化。在转基因小鼠模型中，7-hydroxy MTX的消除与多药转运体Abcc2、Abcc3和Abcg2有关。当所有三种转运体都缺失时，它和母体化合物迅速积累含有甲氨蝶呤的制剂已用于治疗癌症、自身免疫性疾病、异位妊娠和诱导药物流产。在癌症治疗期间，为了避免毒性，已经描述了各种方法来量化7-hydroxy MTX水平。
